Following completion of disease-model mouse studies and additional validation in non-human primates, the IND filing target for MDL-101 was made more specific, shifting from the end of 2026 to the first quarter of 2027. In FY2026-Q3, we will look for tangible progress in GLP toxicology studies, AAV GMP manufacturing and clinical trial readiness, and whether the target timing is delayed again.

For the additional GLP toxicology studies, will the start of dosing during FY2026-Q3 be disclosed as having taken place?
Will AAV GMP manufacturing be described as having started in FY2026-Q3?
Will the IND filing target remain unchanged at “end-2026 to first quarter of 2027”?
Will the assumed patient count for the initial clinical trial change from a total of 6 patients, comprising 3 at the low dose and 3 at the high dose?
Will the FDA filing, selection of trial sites and principal investigator, and status of finalising the clinical trial protocol be disclosed in specific terms?
The company's ongoing themes and initiatives this watchpoint touches on.
The development timeline for MDL-101 has been revised several times in the past. The target had previously been to file the IND in mid-2026, but it was most recently changed from the end of 2026 to the first quarter of 2027 as the company decided to prioritise additional nonclinical evaluation, CMC, analytical systems and optimisation of clinical trial readiness. This appears to represent a concrete shift toward prioritising readiness over the schedule in order to increase the probability of success after clinical transition, rather than merely updating a date.
On the nonclinical side, the company completed IND-enabling studies using disease-model mice and obtained pharmacological efficacy data, including prolonged survival versus the control group, as well as pharmacokinetic data for determining the clinical dose. It also states that a pilot study in non-human primates confirmed vector delivery to muscle tissue, expression of the component factors and increased LAMA1 expression. However, as of FY2026-H1, the quantitative breakdown of these results by sample size, effect size, dose response and long-term expression had not been disclosed. In Q3, it will be important to see whether the results of the additional evaluations are presented as quantitative evidence sufficient to advance to GLP toxicology studies, rather than merely as “confirmation.”
The easiest execution milestones to verify are the start of dosing in the additional GLP toxicology studies scheduled for FY2026-Q3 and the start of AAV GMP manufacturing. The company says that CRO selection and contracting have been completed and that plasmid GMP manufacturing is under way, while AAV GMP manufacturing was scheduled to begin in Q3. In the Q3 results, we will look to distinguish clearly between scheduled, in preparation, started and completed, and to confirm how far the company has progressed with study lots, manufacturing stages and quality evaluation. R&D expenses were ¥900.723 million in FY2026-H1, so if GLP and GMP processes move into full operation, the composition of spending in the second half could also change.
For the clinical design, the assumed initial trial size is 6 patients in total, comprising 3 at the low dose and 3 at the high dose. Whether this patient count, dose selection, dosing method and primary endpoints are maintained in Q3 will be a specific checkpoint for assessing the feasibility of the IND filing. There has been no disclosure that dosing of actual patients has begun. The assessment of clinical-transition risk will change substantially depending on whether Q3 brings disclosure of an IND submission to the FDA, finalisation of trial sites and the principal investigator, or another postponement of the filing date.
Whether the company can move from obtaining nonclinical data to executing GLP and GMP work will determine the credibility of the IND target of end-2026 to the first quarter of 2027.
